Evelopment of an in vitro model of the epithelial-mesenchymal transition in luminal breast cancer

Cell lines derived from luminal breast tumours will be cultured at varying seeding densities and analysed for signs of epithelial-mesenchymal transition (EMT). Initially this will consist of using western blotting analysis to detect any changes in the expression of the cell-cell adhesion protein E-cadherin, and later other markers of EMT. E-cadherin is a key marker of the epithelial phenotype and is frequently lost in cancers correlating with tumour grade, and negatively with aggressiveness and prognosis. Further study will be made using immunofluorescence to determine the subcellular localisation of E-cadherin (and other markers of EMT as appropriate).
